Kinetic determination of serum sorbitol dehydrogenase activity with a centrifugal analyzer.
We describe a mechanized method for centrifugal analyzer determination of sorbitol dehydrogenase in serum, based on conversion of D-fructose to sorbitol with simultaneous oxidation of NADH, in triethanolamine buffer at pH 7.4 and 30 degrees C. The standard curve for this assay is linear to 200 U of activity per liter of serum. The mean within-run precision (CV) of the assay is 0.8%. Results cor...
متن کاملSorbitol dehydrogenase is a cytosolic protein required for sorbitol metabolism in Arabidopsis thaliana.
Sorbitol is converted to fructose in Rosaceae species by SORBITOL DEHYDROGENASE (SDH, EC 1.1.1.14), especially in sink organs. SDH has also been found in non-Rosaceae species and here we show that the protein encoded by At5g51970 in Arabidopsis thaliana (L.) Heynh. possesses the molecular characteristics of an SDH. Using a green fluorescent protein-tagged version and anti-SDH antisera, we deter...
متن کاملStudies on Succinic Dehydrogenase
About 2 years ago the author reported that the activity of purified preparations of succinic dehydrogenase is enhanced by phosphate buffer (2). This enhancement was dependent upon the phosphate concentration and appeared to be relatively specific for phosphate, since other buffers, such as Tris,l imidazole, histidine, glycylglycine, and arsenate did not induce it (2, 3). Arsenate, in fact, inte...
متن کاملStudies on Succinate Dehydrogenase
1. Succinate dehydrogenase flavocoenzyme (“SD-flavin”), previously shown to be an SOLsubstituted riboflavin derivative containing a tertiary nitrogen homoconjugated to the flavin nucleus, was subjected to further hydrolysis and to reduction under acid conditions. Both conditions resulted in the liberation of 1 mole of histidine per mole of flavin. This proves histidine to be the covalent link b...
متن کاملA fluorimetric method for red blood cell sorbitol dehydrogenase activity.
A new fluorimetric method for the quantification of red blood cell (RBC) sorbitol dehydrogenase is described. It is based on the oxidation of sorbitol to fructose, in presence of NAD+, catalysed by the RBC-sorbitol dehydrogenase. The quantity of NADH formed is then measured in a filter fluorimeter. Comparison with an indirect spectrophotometric assay yielded good correlation; however, the prese...
